The present invention discloses a method for the treatment ofFlaviviridae infection that includes the administration of a 2'-branched nucleoside, or a pharmaceutically acceptable prodrug and/or salt thereof, to a human in need of therapy in combination or alternation with a drug that directly or indirectly induces a mutation in the viral genome at a location other than a mutation of a nucleotide that results in a change from serine to a different amino acid in the highly conserved consensus sequence, XRXSGXXXT, of domain B of the RNA polymerase region, or is associated with such a mutation. The invention also includes a method to detect a mutant strain of Flaviviridae and a method for its treatment.

L'invention concerne une méthode permettant le traitement d'une infection à Flaviviridae, comprenant l~administration d'un nucléoside à ramification en 2~ ou d'un précurseur et/ou d'un sel de ce dernier, à un sujet humain nécessitant un traitement, en combinaison ou en alternance avec un médicament qui produit directement ou indirectement une mutation dans le génome viral dans un emplacement autre que la mutation du nucléotide, donnant lieu à une modification de la sérine en un acide aminé différent dans la séquence consensus hautement conservée XRXSGXXXT du domaine B de la région ARN polymérase, ou est associé à une telle mutation. L'invention concerne en outre une méthode permettant la détection d'une souche mutante de Flaviviridae, et une méthode permettant de traiter ce dernier.
