Novel 2-phenyl indoles are disclosed of general formula (I), wherein: R1 represents one of the rests -(CH2)n-S(O)m-R4, -(CH2)n-NR6-SO2-R4, (a), and n' being integers between 4 and 12 and m and m' are 0, 1 or 2; X represents a methylene group, an imino group = NR6, or an oxygen or sulphur atom; R2 and R3 independently of one another represent a hydrogen atom, a C1-C10 alkyl group, a benzyl, alkanoyl or alkanoyloxy, or carbamoyl rest of formula -C(O)R5 or - C(O)NR6R7 or a tetrahydropyranyl group; R4 represents a hydrogen atom, a C1- C10 alkyl, a completely or partially fluorinated alkyl group -(CH2)o- (CF2)pCF3, o and p being independently of one another integers between 0 and 6, an (alkyl)amino or (alkyl)carbamoyl group of formula (CH2)q-Y-NR8R9, q being an integer between 0 and 6 and Y representing a direct bond, a methylene or carbamoyl group (a condition being that q cannot be 0 when Y is a carbonyl group and m is 2), an aryl, aralkyl or heteroaryl group; R5 represents a C1- C10 alkyl group or a C1-C10 alkoxy group, a phenyl or benzyl group; R6, R7, R8 and R9 independently of one another represent a hydrogen atom, a C1-C10 alkyl group, or a benzyl group; and R10 represents a methyl group and R11 represents a hydrogen atom, or R10 and R11 both represent a di-, tri- or tetramethylene bridge which can also have a C-C double bond at any location in the bridge. These novel compounds have strong and selective anti-oestrogen properties and are suitable for use in particular in the treatment of oestrogen-related diseases.
L'invention concerne des 2-phénylindoles de la formule générale (I), dans laquelle R?1¿ représente un des restes: -(CH¿2?)¿n?-S(O)¿m?-R?4¿, -(CH¿2?)¿n?-NR?6¿-SO¿2?-R?4¿, (a), n et n' étant des nombres entiers compris entre 4 et 12, et m et m' valant 0,1 ou 2; X représente un groupe méthylène, un groupe imino = NR?6¿, un atome d'oxygène ou un atome de soufre; R?2¿ et R?3¿ représentent, indépendamment l'un de l'autre, un atome d'hydrogène, un groupe alkyle C¿1?-C¿10?, un reste benzyle, alcanoyle ou alcanoyloxy, ou carbamoyle de la formule: -C(O)R?5¿ ou C(O)NR?6¿R?7¿, ou bien un groupe tétrahydropyranyle; R?4¿ représente un atome d'hydrogène, un groupe alkyle C¿1?-C¿10?, un groupe alkyle complètement ou partiellement fluoré -(CH¿2?)¿o?-(CF¿2?)¿p?CF¿3?, où o et p représentent, indépendamment l'un de l'autre, un nombre entier compris entre 0 et 6, ou bien un groupe (alkyl)amino ou (alkyl)carbamoyle de la formule (CH¿2?)¿q?-Y-NR?8¿R?9¿, où q représente un nombre entier compris entre 0 et 6 et Y représente une liaison directe, ou bien un groupe méthylène ou carbamoyle, à condition que q ne puisse pas valoir 0 lorsque Y représente un groupe carbonyle et m vaut 2, un groupe aryle, aralkyle ou hétéroaryle; R?5¿ représente un groupe alkyleC¿1?-C¿10? ou bien un groupe alcoxy C¿1?-C¿10?, un groupe phényle ou benzyle; R?6¿, R?7¿, R?8¿ et R?9¿ représentent, indépendamment l'un de l'autre, un atome d'hydrogène, un groupe alkyle C¿1?-C¿10?, ou un groupe benzyle; et R?10¿ représente un groupe méthyle et R?11¿ représente un atome d'hydrogène, ou bien R?10¿ et R?11¿ représentent ensemble un pont diméthylène, triméthylène ou tétraméthylène, lequel peut également présenter une liaison double C-C, en n'importe quel point. Ces nouveaux composés ont un fort effet sélectif antiestrogène et peuvent être utilisés pour le traitement de maladies dépendant des estrogènes.
