A process of preparing a 6-O-methyl erythromycin A derivative using a 2'- protected, 9-etheroxime erythromycin A intermediate is provided. A preferred protecting group for the 2'-position is acetyl. 2'- protected, 9-etheroxime erythromycin A derivatives are also provided. Also disclosed is a method for inhibiting quaternary salt formation at the 3' amine without the need for 3'N-protecting groups.

L'invention a pour objet un procédé pour préparer un dérivé de 6-O-méthyle érythromycine A à l'aide d'un intermédiaire qui est une 9-éthéroxime érythromycine A, protégée en 2'. Un groupe de protection préféré pour la position 2' est acétyle. L'invention concerne aussi des dérivés de 9-éthéroxime érythromycine A protégée en 2' ainsi qu'un procédé pour inhiber la formation de sels quaternaires au niveau de l'amine en position 3', sans nécessiter de groupes protecteurs 3'N.
