A class of 4,7-dichlororhodamine compounds useful as fluorescent dyes are disclosed having structure formula (I) wherein R1-R6 are hydrogen, fluorine, chlorine, lower alkyl, lower alkene, lower alkyne, sulfonate, sulfone, amino, amido, nitrile, lower alkoxy, linking group, or combinations thereof, or, when taken together, R1 and R6 is benzo, or, when taken together, R4 and R5 is benzo; Y1-Y4 are hydrogen or lower alkyl, or, when taken together, Y1 and R2 is propano and Y2 and R1 is propano, or, when taken together, Y3 and R3 is propano and Y4 and R4 is propano; and X1-X3 taken separately are selected from the group consisting of hydrogen, chlorine, fluorine, lower alkyl, carboxylate, sulfonic acid, -CH2OH, and a linking group. In another aspect, the invention includes reagents labeled with the 4,7-dichlororhodamine dye compounds, including deoxynucleotides, dideoxynucleotides, and polynucleotides. In an additional aspect, the invention includes methods utilizing such dye compounds and reagents including dideoxy polynucleotide sequencing and fragment analysis methods.
Une classe de composés de 4,7-dichlororhodamine utiles comme colorants fluorescents et représentés par la structure (I) est décrite. Dans cette structure, R1-R6 sont hydrogène, fluor, chlore, alkyle inférieur, alcène inférieur, alcyne inférieur, sulfonate, sulfone, amino, amido, nitrile, alcoxy inférieur, un groupe de liaison ou des combinaisons de ces derniers, ou, pris ensemble, R1 et R6 sont benzo, ou encore, pris ensemble, R4 et R5 sont benzo; Y1-Y4 sont hydrogène ou alkyle inférieur, ou, pris ensemble, Y1 et R4 sont propano et Y2 et R1 sont également propano, ou encore, pris ensemble, Y3 et R3 ainsi que Y4 et R4 sont propano; et X1-X3 sont séparément choisis dans le groupe composé d'hydrogène, de chlore, de fluor, d'alkyle inférieur, de carboxylate, d'acide sulfonique, de -CH2OH et d'un groupe de liaison. Dans un autre aspect, l'invention se rapporte à des réactifs marqués au moyen desdits composés colorants à base de 4,7-dichlororhodamine, y compris des désoxynucléotides, des didésoxynucléotides et des polynucléotides. Dans encore un autre aspect, l'invention concerne des procédés faisant appel à de tels composés colorants et réactifs, comprenant des techniques de séquençage des polynucléotides didésoxy et d'analyse de fragments.
