The present invention is directed to methods of producing conjugates of A.beta. peptide immunogens with protein/polypeptide carrier molecules, which are useful as immunogens, wherein peptide immunogens are conjugated to protein carriers via activated functional groups on amino acid residues of the carrier or of the optionally attached linker molecule, and wherein any unconjugated reactive functional groups on amino acid residues are inactivated via capping, thus retaining the immunological functionality of the carrier molecule, but reducing the propensity for undesirable reactions that could render the conjugate less safe or effective. Furthermore, the invention also relates to such immunogenic products and immunogenic compositions containing such immunogenic products made by such methods.
La présente invention concerne des techniques de production de conjugués immunogènes peptidiques AB avec des molécules porteuses de protéines/polypeptide, qui convienne comme immunogènes, ces immunogènes peptidiques étant conjugués avec des porteurs de protéines via des groupes fonctionnels activés sur des résidus d'acides aminés du porteur ou de la molécule de liaison éventuellement attachée et, n'importe quel groupe fonctionnel réactif non conjugué sur des résidus d'acides aminés sont inactivés par coiffage, retenant ainsi la fonctionnalité immunologique de la molécule porteuses mais réduisant la propension à des réactions indésirables qui pourraient rendre ce conjugué moins sûr ou moins efficace. Par ailleurs, cette invention concerne aussi des produits immunogènes et des compositions immunogènes contenant ces produits immunogènes fabriqués par ces techniques.
