The present invention relates to a novel computer based method for performing genomewise comparison of several organisms, the said computational method involves creation of peptide libraries from protein sequences of several organisms and subsequent comparison leading to identification of conserved invariant peptide motifs, and to this end several invariant peptide motifs have been identified by direct sequence comparison between various bacterial organisms and host genomes without any a priori assumptions, and the present method is useful for identification of potential drug targets and can serve as drug screen for broad-spectrum antibacterial as well as for specific diagnosis of infections, and in addition, for assignment of function to proteins of yet unknown functions with the help of such invariant peptide motif signatures.
La présente invention concerne un nouveau procédé informatique permettant de réaliser des comparaisons de génomes de plusieurs organismes. Ce procédé informatique consiste à créer une bibliothèque de peptides à partir de séquences de protéines de différents organisme et ensuite à comparer ces peptides de façon à identifier les motifs de peptide invariants conservés. A cette fin, plusieurs motifs de peptide invariants ont été identifiés par comparaison de séquences directe entre divers organismes bactériens et des génomes hôtes sans aucun à priori, et ce procédé convient pour identifier des cibles de médicament potentielles et il peut servir d'outil de criblage de médicament pour antibactérien à large spectre, de même qu'il peut servir pour des diagnostics d'infections et pour l'attribution de fonctions jusqu'à présent inconnues à des protéines à l'aide de ces signatures de motif de peptide invariant.
