The present invention relates generally to the field of human genetics, and more specifically to the detection of a specific type of germline mutations in the BRCA1 gene, which will predispose to breast and ovarian cancer. In addition, the invention relates to the molecular genetic mechanism that may have mediated the genesis of these mutations, in particular the role of Alu repetitive DNA elements present in the intronic regions of BRCA1. The invention further relates to somatic mutations of this type in the BRCA1 gene in human breast and ovarian cancer, and their use in the diagnosis and prognosis of human breast and ovarian cancer. The invention more particularly relates to the screening of this type of BRCA1 mutations in human genomic DNA, which are useful for the diagnosis of inherited predisposition to breast and ovarian cancer.
L'invention concerne, de façon générale, le domaine de la génétique humaine et, en particulier, la détection d'un type spécifique de mutations de lignes germinales dans le gène BRCA1, ce qui prédisposera au cancer du sein et des ovaires. Elle concerne, de plus, le mécanisme génétique moléculaire pouvant avoir provoqué la genèse de ces mutations, notamment le rôle joué par des éléments d'A D N à Alu répétitif présents dans les régions introniques de BRCA1. Elle concerne également des mutations somatiques de ce type dans le gène BRCA1 dans le cancer du sein et des ovaires, ainsi que leur utilisation pour le diagnostic et le pronostic du cancer du sein et des ovaires. Elle concerne, encore plus particulièrement, la recherche par criblage de ce type de mutations de BRCA1 dans l'A D N génomique humain, ce qui peut être mis en application afin de diagnostiquer une prédisposition héréditaire au cancer du sein et des ovaires.
