The invention provides an oral drug delivery composition comprising an .alpha.-adreno receptor blocking drug characterised in that the composition is adapted to release a first portion of the drug in the upper gastrointestinal tract and to release a second portion of the drug by sustained release in the terminal ileum and/or the colon. This composition provides a two phase release profile which maintains sufficient and steady plasma levels for therapeutic effect whilst minimising side effects by avoiding a high peak in plasma levels. The sustained release of the second and optionally the first portion of the drug is achieved by a controlled release system such as a hydrophilic gel matrix. The specific release of the second portion of the drug in the colon can be achieved by coating tablets containing the second portion with a pH or redox sensitive coating such as a polymethylmethacrylate.
L'invention concerne une composition médicamenteuse s'administrant oralement et comprenant un agent bloquant du récepteur alpha -adrénergique, caractérisée par le fait que ladite composition est conçue pour libérer une première partie du médicament dans les voies gastro-intestinales supérieures et une deuxième partie du médicament par libération prolongée dans l'iléon terminal et/ou le côlon. La composition présente un profil de libération en deux phases permettant de maintenir des niveaux de plasma suffisants et stables, dans le but de réaliser un effet thérapeutique, tout en minimisant les effets secondaires en évitant des crêtes élevées dans les niveaux de plasma. La libération contrôlée de la deuxième et, éventuellement, de la première partie du médicament s'effectue au moyen d'un système de libération contrôlée, tel qu'une matrice en gel hydrophile. La libération spécifique de la deuxième partie du médicament dans le côlon peut s'effectuer au moyen de comprimés à revêtement contenant la deuxième partie et sensible au pH ou à l'oxidoréduction, tel qu'un polyméthylméthacrylate.
