The invention provides promoter specific and promoter non-specific screening assays for identifying an inhibitor of a pathogenic nucleic acid polymerase activity, e.g., an RNA polymerase derived from a phatogenic infectious organism such as a bacterium, protozoan or fungus. Generally, the methods involve forming a mixture of nucleotide, a polynucleotide template, a pathogenic polymerase, and a candidate inhibitor of polymerase activity, where at least one of the nucleotide comprises a detectable label. In a preferred embodiment, a membrane apparatus (10) having two filters extending transversely across a well, and positioned between the top, tube containing half (11) of the apparatus (10) and the bottom, drip director containing half (15) of the apparatus (10). The top filter (13) is initially hydrophobic and capable of retaining liquid media and is rendered water permeable by contacting it with an organic solvent. The bottom filter (14) is hydrophilic water permeable and efficiently passes liquid media while retaining micro- beads during vacuum filtration.
L'invention porte sur des épreuves de sélection de promoteurs spécifique et non spécifique, visant à identifier un agent inhibiteur de l'activité pathogène d'une polymérase d'acide nucléique, une ARN-polymérase dérivée d'un organisme infectieux pathogène, par exemple, comme une bactérie, un protozoaire ou un micromycète. En règle générale, ce procédé suppose la constitution d'un mélange comprenant des nucléotides, une matrice de polynucléotide, une polymérase pathogène et un agent candidat inhibiteur de l'activité de la polymérase, dans lequel au moins l'un des nucléotides comporte une marque perceptible. Dans une réalisation préférée, on utilise un appareil à membrane (10) pourvu de deux filtres disposés transversalement dans un puits, et situés entre le haut, soit le tube constituant une moitié (11) de l'appareil (10), et le bas, soit le régulateur d'écoulement constituant l'autre moitié (15) de l'appareil (10). Le filtre supérieur (13), qui, au départ est hydrophobe et capable de retenir des milieux liquides, est rendu perméable à l'eau après avoir été mis en contact avec un solvant organique. Le filtre inférieur (14), hydrophile et perméable à l'eau, laisse bien passer des milieux liquides tout en retenant des microparticules durant la filtration sous vide.
