The present invention relates to a method for detecting substances specifically inhibiting a type III secretion mechanism and functions of the type III secretory proteins, within short time and large amounts thereof, without depending upon animal infectious experiments. Namely it relates to the method for detection of a type III secretory mechanism inhibitor comprising mixing a bacterium having the type III secretory mechanism and an erythrocyte suspension, adding the type III secretory mechanism inhibitor thereto, and detecting changes in the thus formed hemolytic activity. The method for detecting substances can be treated large amount of samples within short time by exhibiting the substances inhibiting the type III secretion mechanism or the functions of the type III secretory proteins as numerical index of the hemolytic activity of erythrocytes. Consequently , the present invention is useful for development of drugs.
L'invention concerne un procédé permettant de détecter un composant qui inhibe de manière spécifique le mécanisme de sécrétion du type III et la fonction d'une protéine sécrétoire de type III sécrétée par ce mécanisme, en peu de temps et avec un débit élevé, sans qu'il soit nécessaire de recourir à des expériences impliquant l'infection d'animaux. Ce procédé permettant de détecter un inhibiteur du mécanisme de sécrétion de type III est caractérisé par les étapes suivantes : on mélange une bactérie présentant le mécanisme de sécrétion de type III avec une suspension d'érythrocytes, on ajoute un inhibiteur de ce mécanisme de sécrétion du type III à ce mélange, et on détecte une modification de l'activité hémolytique causée par cet inhibiteur. Ce procédé permet de traiter un grand nombre d'échantillons en un intervalle de temps bref et permet d'exprimer numériquement un composant inhibant le mécanisme de sécrétion de type III d'une bactérie ou la fonction d'une protéine sécrétoire de type III, à partir de l'indication fournie par l'activité hémolytique des érythrocytes. Ces caractéristiques rendent ce procédé avantageux, par exemple pour la mise au point de médicaments.
