A method is described to assay for protein interactions in living cells, e.g. by the introduction of two heterologous conjugates into the cell. The method uses the measurement of cellular distribution of a detectable component (e.g. a GFP-labelled fluorescent probe) to indicate the presence or absence of an interaction between that component and a second component of interest. The method uses the knowledge that certain components can be stimulated to redistribute within the cell to defined locations. Inducible redistribution systems make it possible to determine if specific interactions occur between components. Inducible systems are described where it is demonstrated that the redistribution stimuli are essentially "null", in that they affect no other system in the cell during the assay period, other than the component whose redistribution can be induced. Also described is an extraction buffer which is useful in high throughput screening for drugs which affect the intracellular distribution of intracellular components. The extraction buffer comprises a cellular fixation agent and cellular permeabilisation agent. Optimising the composition of the extraction buffer and its application to various cell types is described.

La présente invention concerne une technique permettant d'analyser des interactions de protéines dans des cellules vivantes. Cette technique utilise la mesure de distribution cellulaire d'un composant détectable de façon à indiquer la présence ou l'absence d'une interaction entre ce composant et un deuxième composant étudié. Cette technique se fonde sur la connaissance du fait que certains composants peuvent être stimulés pour se redistribuer dans la cellule à des localisation définies. Des système de redistribution inductibles permettent de déterminer si des interactions spécifiques surviennent entre composants. Cette invention concerne aussi des systèmes inductibles dans lesquels on démontre que les stimuli de redistribution sont sensiblement <=nuls>=, en ce sens qu'ils n'affectent aucun autre système dans la cellule durant la période d'analyse, sinon le composant dont la redistribution peut être induite. Cette invention concerne aussi un tampon d'extraction qui convient pour le criblage à haut rendement des médicaments qui ont une action sur la distribution intracellulaire des composants intracellulaires. Ce tampon d'extraction comprend un agent de fixation cellulaire et un agent de perméabilisation cellulaire. Cette invention concerne enfin l'optimisation de la composition du tampon d'extraction et son application à divers types de cellule.
