The invention is a method for genomic subtractive hybridization. Specific nucleic acid sequences are removed from a sample of nucleic acid sequences by specifically hybridizing the sequences to a complementary nucleic acid sequence bound to a target molecule such as biotin. The target molecule is then contacted with a binding partner such as avidin and separated from the sample of nucleic acid sequences. As the target is separated from the sample the hybridized nucleic acid sequences are also removed from the sample. The method preferably involves the removal of repetitive nucleic acid sequences from a nucleic acid sample to generate a library of probes that are substantially free of repetitive nucleic acid sequences.
L'invention concerne une technique d'hybridation soustractive. On retire des séquences d'acides nucléiques spécifiques sont retirées d'un échantillon de séquences d'acides nucléiques en hybridant spécifiquement lesdites séquences avec une séquence d'acides nucléiques complémentaire liée à une molécule cible telle que la biotine. Puis on met la molécule cible en présence d'un partenaire de liaison tel que l'avidine et on la sépare de l'échantillon de séquences d'acides nucléiques. En séparant la cible de l'échantillon, on retire en même temps les séquences d'acides nucléiques. Selon la technique, on retire de préférence de l'échantillon des séquences d'acides nucléiques répétitives, afin de générer une banque de sondes sensiblement dépourvue de séquences répétitives d'acides nucléiques.
