A method for high loading of vesicles with biopolymeric substances functional in human biochemical or physiological systems derived from any source, said substances preferably are enzymes proenzymes, cofactors submicron particles such as virions, growth factors, cytokines, robosomes, hepatitis B surface antigen, oligo- and polynucleotides, antibodies and/or antigens, said vesicles formed by amphiphatic substances comprising the steps of a) mixing lipids suitable for forming lipid vesicles in water-immiscible organic solvents, alternatively, dried lipids or lipid mixture can be used in any form (powder, granular, etc.) directly; b) removing the solvent in presence of a solid support; c) taking up the product of step b) into a solution of the substances to be encapsulated in a physiologically compatible solution; d) adding an organic solvent having solubilizing or disposing properties, as well as e) drying the fraction obtained in step d) under conditions retaining the function of the substances to be encapsulated.
Méthode de chargement élevé de vésicules par des substances biopolymères jouant un rôle dans les systèmes biochimiques ou physiologiques humains et provenant de n'importe quelle source. Lesdites substances sont de préférence des enzymes, des proenzymes, des cofacteurs, des particules submicrométriques telles que des virions, des facteurs de croissance, des cytokines, des ribosomes, l'antigène d'enveloppe du virus de l'hépatite B, des oligo- et polynucléotides, des anticorps et/ou des antigènes, lesdites vésicules étant formées par des substances amphiphatiques, en procédant selon les étapes suivantes: a) mélange des lipides appropriés à la formation de vésicules lipidiques dans des solvants organiques non-missibles à l'eau; autrement, des lipides secs ou un mélange lipidique peuvent être utilisés directement sous n'importe quelle forme (pulvérulente, granulaire, etc.); b) extraction du solvant en présence d'un support solide; c) prise du produit de l'étape b) dans une solution de substances devant être encapsulées dans une solution physiologiquement compatible; d) adjonction d'un solvant organique présentant des propriétés de solubilisation ou d'élimination; ainsi que e) assèchement de la fraction obtenue de l'étape d) dans des conditions assurant la préservation de la fonction des substances devant être encapsulées.
