The present invention provides a method and a multi-gene assembly vector system having an acceptor vector and two donor vectors for effective transfer of multiple genes into a single vector. Using the method and the vector system disclosed in this invention, multiple cycles of DNA recombination based on a recombination system can be repeatedly carried out by alternate use of the two donor vectors together with the acceptor vector for recombination. The vector system and the method are used to construct any genetic engineering vector that contains multiple genes or DNA fragments of interest for transfer and expression of the genes in different cells, tissues, and organisms.
