The present invention relates to a method for preparing Langerhans cells (LC), or dermal/interstitial dendritic cells (IDC) or LCs and IDCs from CD 14+ monocytes stemming from peripheral circulatory blood of a living being, and notably of a human being, comprising differentiation of CD 14+ monocytes into LCs, or into IDCs, or into LCs and IDCs by putting the CD 14+ monocytes in the presence of a cell environment comprising epithelial cells, such as keratinocytes, and/or mesenchymatous cells such as dermal fibroblasts. The present invention also relates to cell or tissue models comprising at least LCs and/or IDCs, and optionally macrophages and endothelial cells, able to be obtained according to this method, and to their uses.
La présente invention concerne un procédé de production de cellules de Langerhans (LC) ou des cellules dendritiques dermiques/interstitielles (IDC) ou des LC et IDC à partir de monocytes CD14+ provenant du sang circulatoire périphérique d'un être vivant, et notamment d'un être humain, comprenant la différenciation de monocytes CD14+ en LC, ou en IDC, ou en IDC et LC en mettant les monocytes CD14+ en présence d'un environnement cellulaire comprenant des cellules épithéliales, telles que des kératinocytes et/ou des cellules mésenchymateuses sous forme de fibroblastes dermiques. La présente invention concerne également des modèles cellulaires ou tissulaires comportant au moins des LC et/ou des IDC, et éventuellement des macrophages et des cellules endothéliales susceptibles d'être obtenus par ces procédés, ainsi que leurs utilisations.
