A process for preparing in a good yield optically active erythro-3-amino-2- hydroxybutyric esters having a high purity and serving as an important intermediate of pharmaceuticals, particularly HIV protease inhibitors. The process is characterized by oxidizing the hydroxyl group at the 2-position of a 3-amino-2-hydroxybutyric ester, with the 3-position being optically active, represented by formula (I) (wherein R1 represents a phenyl or cyclohexyl group, R2 represents a protective group, R3 represents a protective group, and the configuration represented by *1 is S-configuration or R-configuration) and then conducting erythro-selective reduction using an aluminum alkoxide.

Cette invention concerne un procédé de préparation de grandes quantités d'esters érythro-3-amido-2-hydroxybutyrique actifs sur le plan optique, lesquels possèdent une grande pureté et peuvent être utilisés en qualité d'intermédiaires importants pour des produits pharmaceutiques, notamment pour des inhibiteurs de protéase du VIH. Ce procédé consiste à oxyder le groupe hydroxyle à la position 2 d'un ester 3-amino-2-hydroxybutyrique dont la position 3 reste active sur le plan optique. Cet ester correspond à la formule (I) où R?1¿ représente un groupe cyclohexyle ou phényle, et R?2¿ et R?3¿ représentent chacun un groupe protecteur. La configuration représentée par *1 consiste en une configuration S ou en une configuration R. Ce procédé consiste enfin à effectuer une réduction à sélection érythro à l'aide d'un alcoxyde d'aluminium.
