A method for the purification and separation of nucleic acid mixtures by chromatography comprising adsorbing the nucleic acids to be separated and purified from a solution with a high concen- tration of salts (ionic strength) and/or a high concentration of alcohol on a substrate and subsequent desorbing from said sub- strate by means of a solution with lower concentration of salts (ionic strength) characterized in that said nucleic acid mixture - is adsorbed from an aqueous adsorption solution with a high concentration of salts (ionic strength) and with 1 to 50% by volume of an aliphatic alcohol having a chain length of C1-C5 and/or polyethylene glycol (PEG) and/or hydrophobic inorganic and/or organic polymers and/or organic acid, such as trichloroacetic acid (TCA), on a porous or non-porous mineral substrate of metal oxides and/or mixed metal oxides, silica gel, materials principally consisting of glass, alumina, zeolites, titanium dioxide, zirconium diox- ide; - is optionally washed with a washing solution, and thereaf- ter - is eluted with a solution with a lower concentration of salts (ionic strength), and the nucleic acid or nucleic acid fraction obtained is collected.

L'invention concerne un procédé de purification et de séparation chromatographiques de mélanges d'acides nucléiques par adsorption des acides nucléiques à purifier et à séparer, contenus dans une solution qui comporte une forte concentration en sels (force ionique) et/ou une forte concentration en alcools sur un support. Cette opération est suivie d'une désorption depuis le support au moyen d'une solution dont la concentration en sels (force ionique) est plus faible. Ce procédé se caractérise en ce que le mélange d'acides nucléiques est adsorbé sur un support minéral poreux ou non poreux comprenant des oxydes métalliques et/ou des oxydes métalliques mixtes, du gel de silice, des matériaux essentiellement à base de verre, de l'oxyde d'aluminium, des zéolithes, du dioxyde de titane, du dioxyde de zirconium provenant d'une solution aqueuse d'adsorption à forte concentration en sels (force ionique), et comportant entre 1 et 50 % en volume d'alcool aliphatique d'une longueur de chaîne de C1 - C5 et/ou du polyéthylène-glycol (PEG) et/ou des polymères inorganiques et/ou organiques hydrophobes et/ou des acides organiques, comme l'acide trichloracétique (TCA). Ce mélange est éventuellement lavé avec une solution de lavage et est ensuite élué avec une solution dont la concentration en sels (force ionique) est plus faible, et l'acide nucléique ou la fraction d'acide nucléique obtenu(e) est receuilli(e).
