RBC and platelet (Plt) alloimmunization requires antigen-matched blood to avoid adverse transfusion reactions. Some blood collection facilities use unregulated Abs to reduce the cost of mass screening, and later confirm the phenotype with government approved reagents. Alternatively, RBC and Plt antigens can be screened by virtue of their associated single nucleotide polymorphisms (SNPs). We developed a multiplex PCR-oligonucleotide extension assay using the GenomeLab SNPStream platform to genotype blood for a plurality of blood group antigen-associated SNPs, including but not limited to: RhD (2), RhC/c, RhE/e, S/s, K/k, Kpa/b, Fya/b, FY0, Jka/b, Dia/b, and HPA-1a/b.
L'allo-immunisation RBC et de plaque sanguine nécessite du sang correspondant à un antigène afin d'éviter des réactions négatives à la transfusion. Certaines installations de collecte de sang utilisent un ABS non réglé pour réduire le coût du dépistage en masse et pour confirmer par la suite le phénotype avec des réactifs autorisés. Selon une variante, les antigènes RBC et de plaquette sanguine peuvent être dépistés au moyen de leurs polymorphismes nucléotidiques simples associés (SNP). Un essai d'extension PCR-oligonucléotide multiplex a été mis au point au moyen d'une plate-forme GenomeLab SNPStream afin d'effectuer le génotype du sang pou une pluralité de SNP associé à un antigène du groupe sanguin, entre autres: RhD (2), RhC/c, RhE/e, S/s, K/k, Kp?a/b¿, Fya/b, FY0, Jk?a/b¿, Di?a/b¿, et HPA-1a/b.
