A method of estimating sepsis risk in an individual with infection comprises a step of assaying a biological sample from the individual for an IL-2 or IL-7 mRNA value, and correlating the mRNA value with sepsis risk. The IL-2 and IL-7 mRNA values are quantified by absolute quantification of mRNA copy number, wherein the copy numbers are normalised to a house keeping gene and corrected against a calibration curve for serial dilutions of the IL-2 and IL-7 cDNA. The method generally involves a step of assaying a biological sample from the individual for IL-2 and/or IL-7 mRNA values, optionally in combination with mRNA values for other cytokines, and correlating a sum or difference of the values with sepsis risk using a regression analysis curve against outcome.
La présente invention concerne un procédé destimation du risque de sepsie chez un individu ayant une infection comprenant une étape consistant à doser un échantillon biologique provenant dun individu à la recherche dune valeur de l'ARNm de lIL-2 ou de lIL-7, et à corréler la valeur de l'ARNm avec le risque de sepsie. Les valeurs de lARNm de lIL-2 et de lIL-7 sont quantifiées par une quantification absolue du nombre de copies de lARNm, le nombre de copies étant normalisé à un gène domestique et corrigé en fonction dune courbe détalonnage construite à laide de dilutions en série des ADNc de lIL-2 et de lIL-7. Le procédé comprend généralement une étape consistant à doser un échantillon biologique provenant de l'individu pour déterminer les valeurs de l'ARNm de l'IL-2 et/ou de l'IL-7, éventuellement en association avec des valeurs de lARNm pour dautres cytokines, et à corréler une somme ou une différence des valeurs avec le risque de sepsie en utilisant une courbe danalyse de régression en fonction du résultat.
