The present invention relates generally to a method of identifying differences between nucleic acid molecules and provides a method capable of identifying a variation in a nucleotide sequence in nucleic acid molecules based on the selectivity of oxidizing agents towards mismatched or unmatched bases in a nucleic acid duplex. The method of the present invention enables the detection of nucleotide variations, such as resulting from base changes (mutations and polymorphisms) in target nucleic acid heteroduplex molecules derived from wildtype and mutant homoduplexes

L'invention porte d'une manière générale sur un procédé d'identification de différences entre des molécules d'acides nucléiques par identification d'une modification de la séquence nucléotidique de molécules d'acides nucléiques se basant sur la sélectivité d'agents oxydants envers des bases mal ou non appariées de duplex d'acides nucléiques. Ledit procédé permet de détecter des modifications des nucléotides telles que celles résultant des modifications de base (mutations et polymorphisme) dans des molécules cibles d'acides nucléiques hétéroduplex dérivant de types sauvages ou de mutants homoduplex.
