The present invention is a method of preparing affinity ligands, which method comprises to provide a cyclic scaffold comprising a thiol group, a carbonyl group and an amine group; to derivatise the amine group of said scaffold with an electrophile that carries either a functionality capable of affinity interaction or a functionality capable of a second interaction with a target molecule; and to open up the derivatised scaffold and add an amine that carries either a functionality capable of a second interaction or a functionality capable of affinity interaction with a target molecule. The method provides provide affinity ligands, which are capable of affinity interaction as well as a second kind of interaction.
L'invention concerne un procédé permettant la préparation de ligands d'affinité. Ce procédé consiste à prépare un squelette cyclique comprenant un groupe thio, un groupe carbonyle et un groupe amine, à modifier le groupe amine dudit squelette avec un électrophile pour former un dérivé, cet électrophile étant porteur soit d'un groupement fonctionnel capable d'une interaction d'affinité, soit d'un groupement fonctionnel capable d'une seconde interaction avec une molécule cible, et à ouvrir le squelette modifié et à ajouter une amine qui porte soit un groupement fonctionnel capable d'une seconde interaction, soit un groupement fonctionnel capable d'une interaction d'affinité avec la molécule cible. Ce procédé permet d'obtenir des ligands d'affinité qui permettent une interaction d'affinité ainsi qu'un second type d'interaction.
