A method for producing IgG from plasma for medical applications, comprising at least: (i') removal of albumin resulting in an IgG fraction, (ii') purifying IgG from an IgG fraction, which is derived from the IgG fraction obtained in step (i'), by adsorbing IgG to a cation exchanger and collecting the adsorbed IgG fraction, and (iii') virus inactivation in an IgG fraction derived from the IgG fraction collected in step (ii'). The method is characterized in; (I) concentrating the IgG fraction obtained in step (i'), (II) adjusting pH to 4.plusmnØ1 in the IgG fraction released from the cation exchanger in step (ii'), and preferably maintaining the pH below 6.0 during the remaining steps of the method; and (III) carrying out the virus inactivation (step iii') by using chemicals at a temperature of 30 ~C .plusmn. 2 ~C for at least 4 hours. Anticomplementary activity is typically below 1 CH50/mg immunoglobulin.
L'invention concerne un procédé de production d'IgG à partir de plasma à des fins médicales. Ce procédé comporte au moins les étapes consistant à : 1) éliminer l'albumine pour obtenir une fraction d'IgG ; 2) purifier l'IgG d'une fraction d'IgG dérivée de la fraction d'IgG obtenue à l'étape 1, par adsorption des IgG vers un échangeur de cations et recueil de la fraction d'IgG adsorbée ; et 3) inactiver les virus dans une fraction d'IgG dérivée de la fraction d'IgG collectée à l'étape 2. Le procédé est caractérisé par (I) la concentration de la fraction d'IgG obtenue à l'étape 1 ; (II) le réglage du pH à 4.plusmn.0,1 dans la fraction d'IgG sortant de l'échangeur de cations à l'étape 2, et de préférence le maintien du pH à une valeur inférieure à 6,0 au cours des étapes suivantes du procédé ; et (III) l'inactivation des virus (étape 3) mise en oeuvre à l'aide de produits chimiques à une température de 30 ·C .plusmn. 2 ·C pendant au moins quatre heures. L'activité anticomplémentaire est généralement inférieure à 1 CH¿50?/mg d'immunoglobulines.
