A method of producing male or female sterile plants comprising the steps of transforming plant material with a polynucleotide which encodes at least one enzyme which reacts with a non-phytotoxic substance to produce a phytotoxic one, and regenerating the thus transformed material into a plant, wherein the said non-phytotoxic substance is applied to the plant up to the time of male or female gamete formation and/ or maturation, so that the non-phytotoxic substance provides for the production of a phytotoxic one which selectively prevents the formation of or otherwise renders the said gametes non- functional, wherein the enzyme is expressed preferentially in either male, or female reproductive structures and the non-phytotoxic substance is a D-alpha amino acid, or a peptide derivative of a non-protein D-alpha amino acid, characterised in that the enzyme is a mutant D-amino acid oxidase, obtainable from Rhodotorula gracilis, which oxidase comprises a lysine at position 58 rather than a phenylalanine in the wild type sequence.

L'invention concerne un procédé de production sélective de plantes stériles mâles ou femelles, comprenant les étapes suivantes : transformation du matériau de la plante par un polynucléotide qui code au moins une enzyme réagissant avec une substance non-phytotoxique pour produire une substance phytotoxique, et régénération du matériau ainsi transformé en une plante, ladite substance non-phytotoxique étant appliquée à la plante au moment de la formation de gamète mâle ou femelle et/ou de la maturation, de telle façon que la substance non-phytotoxique assure la production d'une substance phytotoxique empêchant sélectivement la formation desdites gamètes ou bien rendant celles-ci non fonctionnelles, l'enzyme étant exprimée, de préférence, dans des structures reproductives mâles ou femelles. La substance non-phytotoxique est un aminoacide D-alpha, ou un dérivé de peptide d'un aminoacide D alpha non-protéine. L'invention est caractérisée en ce que l'enzyme est une oxydase aminoacide D mutante, obtenue à partir de Rhodotorula gracilis, et en ce que ladite oxydase comprend une lysine en position 58 plutôt qu'une phénylalanine dans la séquence de type sauvage.
