The present invention relates to a humanized, non-human mammal model, preferably a humanized mouse model, with an engrafted portion of human skin having hypersensitivity to ultra violet (UV) light, and a method for preparing such non-human mammal model and its use for studying acute and long term effects on human skin by exposure to UV light and for testing topically or systematically applied or administered substances for their capability to prevent, repair and/or cure damages of such exposed human skin. This mouse model comprises an engrafted portion of human skin based on skin cells of a human Xeroderma pigmentosum (XP) or Gorlin' s Syndrome (GS) patient. XP and GS are rare human autosomal disorders characterized clinically by hypersensitivity to UV light, especially to UVB rays, and high predisposition for developing skin cancers on sunlight exposed skin areas.

L'invention concerne un modèle de mammifère non humain, humanisé, de préférence, un modèle de souris humanisé comprenant une partie de peau humaine greffée hypersensible à la lumière ultraviolette (U.V.), un procédé de préparation de ce modèle non humain, et son utilisation pour étudier les effets aigus et à long terme de l'exposition de la peau humaine aux U.V., et pour tester des substances appliquées ou administrées de manière topique ou systématique permettant d'empêcher, de réparer et/ou de soigner les dommages causés par une exposition de la peau humaine aux U.V. Ledit modèle de souris comprend: une partie de peau humaine greffée à base de cellules cutanées provenant d'un patient humain atteint de Xeroderma pigmentosum (XP) ou du syndrome de Gorlin (GS). XP et GS sont des troubles autosomaux humains rares caractérisés cliniquement par une hypersensibilité à la lumière ultraviolette, notamment aux rayons U.V.B, et une prédisposition élevée à développer des cancers de la peau sur des zones cutanées exposées à la lumière solaire.
