This invention provides an isolated strain of Hepatitis B virus designated Human Hepatitis B Virus Surface Antigen-'S'-133 Oon Strain (Methionine to Threonine) which constituent viral genome is deposited under Accession Nos. P97121501, P97121502 and P97121503 with the European Collection of Cell Culture on 15th December 1997. This invention also provides an isolated nucleic acid encoding a polypeptide which is a mutant major surface antigen of a strain of hepatitis B virus, such polypeptide having an amino acid sequence which differs from the amino acid sequence of a major surface antigen of a wild type hepatitis B virus in that the amino acid at position number 133 of such polypeptide is a threonine rather than a methionine. This invention also provides an isolated nucleic acid which encodes a peptide, wherein the peptide is encoded by a nucleic acid molecule comprising nucleotides 527 through 595 of SEQ. ID. No. 1 and the purified peptide. This invention also provides various methods of using the disclosed isolated nucleic acid and peptides. This invention further provides various uses of the disclosed isolated nucleic acid, polypeptides and peptides, and antibodies.

L'invention concerne une souche isolée de l'hépatite B appelée souche d'antigène-'S'-133 Oon de surface de l'hépatite B humaine (de la méthionine à la thréonine) dont le génome viral constitutif a été breveté sous les n DEG P97121501, P97121502 et P97121503 avec l'ensemble européen de culture cellulaire le 15 décembre 1997. L'invention concerne aussi un acide nucléique isolé codant pour un polypeptide qui est un antigène de surface mutant principal d'une souche de l'hépatite B, ce polypeptide ayant une séquence d'aminoacides différente de la séquence d'un antigène de surface principal d'un type sauvage du virus de l'hépatite B. La différence vient de ce que l'aminoacide en position n DEG 133 de ce polypeptide est une thréonine au lieu d'une méthionine. Cette invention concerne également un acide nucléique isolé codant pour un peptide, le peptide étant codé par une molécule d'acide nucléique renfermant des nucléotides de 527 à 595 de SEQ. ID No. 1 et le peptide purifié. L'invention concerne en outre plusieurs procédés d'utilisation de l'acide nucléique isolé et de peptides. L'invention concerne finalement différentes applications de l'acide nucléique isolé, des polypeptides et des peptides, ainsi que des anticorps.
