An isolated NBMPR-insensitive equilibrative nucleoside transport protein (iENTP) and the nucleic acid encoding it is disclosed. The iENTP can be used in screening assays to identify both natural nucloside permeants and/or inhibitors and analogs thereof. In addition, transfected or transduced cell lines are disclosed which use the iENTP as the sole nucleoside transport protein. Methods of employing such cell lines for drug screening are also included. Furthermore methods of using hematopoietic stem cells transduced with an iENTP in a chemotherapy protocol is also described. In addition, methods of using these cells to selectively express a heterologous gene for gene therapy is disclosed.
L'invention a trait à une protéine de transport de nucléoside (iENTP), équilibratrice et insensible au NBMPR, ainsi qu'à l'acide nucléique la codant. On peut utiliser cette protéine dans des criblages aux fins de l'identification à la fois de nucléosides, d'origine naturelle, transmenbranaires et susceptibles de se fixer à la protéine iENTP (appelés permeants) et/ou d'inhibiteurs et d'analogues de ceux-ci. Cette invention concerne également des lignées cellulaires transfectées ou transduites utilisant la protéine iENTP comme unique protéine de transport de nucléoside, des méthodes d'application de ces lignées cellulaires pour des criblages de médicaments, des méthodes d'utilisation de lignées cellulaires hématopoïétiques transduites par la protéine iENTP dans le cadre d'un protocole de chimiothérapie ainsi que des méthodes d'utilisation de ces cellules aux fins d'une expression sélective d'un gène hétérologue pour thérapie génique.
