Disclosed herein are a pharmaceutical composition for inhibiting the growth or metastasis of cholangiocarcinoiria, comprising a LlCAM activity inhibitor or expression suppressor and a treatment method using the composition. This is based on the finding that LlCAM is overexpressed on cholangiocarcinoma and plays an important role in the growth and metastasis of cholangiocarcinoma and the mortality of cholangiocarcinoma patients increases as the expression rate of LlCAM increases. Also, antibodies inhibitory of the activity of LlCAM, or siRNAs suppressing the expression of LlCAM, are found to reduce the growth and invasion of cholangiocarcinoma cells. Mouse monoclonal antibodies, recognizing the LlCAM protein on the cholangiocarcinoma cell surface and binding specifically to cholangiocarcinoma tissues, or siRNAs, antisense oligonucleotides or shRNAs, may be useful in the treatment of cholangiocarcinoma by inhibiting the growth, invasion and migration of cholangiocarcinoma cell.
Composition pharmaceutique inhibant la croissance métastasique d'un cholangiocarcinome, qui renferme un inhibiteur d'activité LICAM ou un suppresseur d'expression LICAM, et procédé de traitement faisant appel à la composition. On se fonde sur la découverte selon laquelle on est en présence de surexpression de LICAM sur un cholangiocarcinome, ce phénomène jouant un rôle important dans la croissance et la métastase du cholangiocarcinome et la mortalité correspondante augmentant à mesure que le taux d'expression de l'activité LICAM augmente. En outre, on décrit des anticorps inhibant l'activité LICAM, ou des ARNsi supprimant l'expression LICAM, pour réduire la croissance et l'invasion des cellules correspondantes. Par ailleurs, des anticorps monoclonaux de souris, reconnaissant la surface de ces cellules et se liant spécifiquement aux tissus de cholangiocarcinome, ou des ARNsi, des oligonucléotides antisens ou des ARNsh, peuvent être utiles dans le traitement du cholangiocarcinome, par inhibition de la croissance, de l'invasion et de la migration des cellules considérées.
