Disclosed is a protein complex, comprising a physiologically active polypeptide, a dimeric protein and a non-peptidyl polymer having three functional ends(3-arm), with the linkage of both the physiologically active polypeptide and the dimeric protein to the 3-arm non-peptidyl polymer via respective covalent bonds. The protein complex guarantees the long acting activity and biostability of a physiologically active polypeptide. Having the ability to maintain the bioactivity of physiologically active polypeptides or peptides highly and to significantly improve the serum half life of the polypeptides or peptides, the protein complex can be applied to the development of sustained release formulations of various physiologically active polypeptide drugs. Also, it utilizes raw materials including the physiologically active polypeptides without significant loss, thereby increasing the production yield. Further, it can be easily purified.
La présente invention concerne un complexe de protéine, comprenant un polypeptide physiologiquement actif, une protéine dimérique et un polymère non peptidylique ayant trois extrémités fonctionnelles (3 bras), la liaison du polypeptide physiologiquement actif et de la protéine dimérique au polymère non-peptidylique à 3 bras étant effectué via des liaisons covalentes respectives. Le complexe de protéine garantit lactivité et la biostabilité durables dun polypeptide physiologiquement actif. Grâce à sa capacité à maintenir fortement la bioactivité de polypeptides ou peptides physiologiquement actifs et daméliorer significativement la demi-vie sérique des polypeptides ou peptides, le complexe de protéine peut être appliqué au développement de formulations à libération prolongée de différents médicaments polypeptidiques physiologiquement actifs. De plus, il utilise des matières premières comprenant les polypeptides physiologiquement actifs sans perte significative, de manière à augmenter le rendement de production. De plus, il peut être aisément purifié.
