A process for preparation of imiquimod comprising oxidation of 1-Isobutyl-1H- imidazo quinoline quinoline (II) afforded 1-Isobutyl-lH- imidazo-[4,5-c]- quinoline-5-N-oxide (III) which is isolated in pure form as its hydrochloride salt (IV) followed by conversion to 4-chloro derivative(V) and conversion to corresponding 4-iodo derivative (VI) which is a new intermediate. This new intermediate is convened to imiquimod (VIII) and purified via its novel maleate salt.

La présente invention a pour objet un procédé de synthèse de l'imiquimod qui comprend l'oxydation de la 1-Isobutyl-1H-imidazo quinoléine (II), aboutissant à la formation de 1-Isobutyl-1H-imidazo-[4,5-c]-quinoléine-5-N-oxyde (III), lequel est isolée sous une forme pure de chlorhydrate (IV), avant d'être converti en dérivé 4-chloro (V), puis en dérivé 4-iodo (VI), qui constitue un nouvel intermédiaire de l'invention. Ce nouvel intermédiaire est alors converti en imiquimod (VIII), puis purifié sous la forme d'un nouveau sel d'acide maléique selon l'invention.
