The present invention relates to a pure form of rapamycin with a total impurity content less than 1.2% ; a process for recovery and purification of rapamycin comprising steps of (a) treating the fermentation broth, extracts or solutions containing rapamycin with water immiscible solvent and concentration; (b) addition of a water miscible solvent to effect separation of impurities present; (c) optionally, binding of the solvent containing the product from step (b) to an inert solid, washing the solid with a base and acid, followed by elution; (d) subjecting the elute from step (c) or the solvent containing the product from step (b) to silica gel chromatography; (e) crystallization of the product obtained from step (d); (f) subjecting a solution of the product from step (e) to hydrophobic interaction or reversed phase chromatography; and (g) re-crystallization to afford rapamycin in substantially pure form.
La présente invention concerne une forme pure de rapamycine avec une teneur totale en impuretés inférieure à 1,2 % ; un procédé de récupération et de purification de la rapamycine comprenant les étapes consistant à (a) traiter le bouillon de fermentation, les extraits ou les solutions contenant la rapamycine avec un solvant non miscible à l'eau et concentrer ; (b) ajouter un solvant miscible à l'eau pour effectuer la séparation des impuretés présentes ; (c) éventuellement, lier le solvant contenant le produit issu de l'étape (b) à un solide inerte, laver le solide avec une base et un acide, ceci suivi d'une élution ; (d) soumettre l'éluat issu de l'étape (c) ou le solvant contenant le produit issu de l'étape (b) à une chromatographie sur gel de silice ; (e) cristalliser le produit obtenu de l'étape (d) ; (f) soumettre une solution du produit issu de l'étape (e) à une interaction hydrophobe ou à une chromatographie en phase inverse ; et (g) recristalliser pour produire la rapamycine sous une forme pratiquement pure.
