The present invention relates to a method for detecting the presence of the abnormal isoform of prion protein (PrPSC) in a urine sample of a subject. The method of the invention comprising the steps of: (a) providing a urine sample of said subject; (b) isolating from said sample all proteins, preferably, isolating proteins having a molecular weight higher than about 8 Kda; (c) optionally, and preferably, subjecting the proteins obtained in step (b) to protease digestion, and isolating from the mixture obtained in step (c) any protease resistant proteins; and (d) detecting the presence of PrPSC in the protease resistant fraction obtained in step (c) by a suitable detection technique.Furthermore, the invention further relates to methods for diagnosing a prion disease in a subject and for screening donors of blood samples for the presence of prion diseases. The invention further provides for a diagnostic kit for diagnosing a prion disease in a subject.
La présente invention concerne une méthode de détection de la présence de l'isoforme anormale de la protéine prion (PrPSC) dans un échantillon d'urine d'un sujet. La méthode de l'invention consiste (a) à obtenir un échantillon d'urine du sujet; (b) à isoler toutes les protéines de l'échantillon et, de préférence, à isoler les protéines dont la masse moléculaire est supérieure à environ 8 Kda; (c) éventuellement et de préférence, à soumettre les protéines obtenues au cours de l'étape (b) à la digestion des protéases, et à isoler du mélange obtenu au cours de l'étape (c) toute protéine résistant aux protéases; et (d) à détecter la présence de PrPSC dans le fragment résistant aux protéases obtenu au cours de l'étape (c) par une technique de détection adaptée. De plus, l'invention concerne des méthodes de diagnostic d'une maladie à prion chez un sujet, et de criblage de donneurs d'échantillons de sang à la recherche de la présence de maladies à prion. L'invention concerne également un kit de diagnostic servant à diagnostiquer une maladie à prion chez un sujet.
