A simple and efficient method for random in vitro mutagenesis and recombination of polynucleotide sequences based on walk-through with chain- elongating molecules and chain terminating molecules e.g. dNTPs/ddNTPs followed by reassembly and amplification is described. The utility of this method for improving protein structure and/or function is demonstrated by creating novel sarcosine oxidase variants and by recombining human placental alkaline phosphates and calf intestinal alkaline phosphatase for better mutants with improved properties.

L'invention concerne un procédé simple et efficace de mutagenèse et de recombinaison aléatoires in vitro de séquences polynucléotidiques sur la base d'une révision structurée avec des molécules d'allongement de chaîne et des molécules de terminaison de chaîne, par exemple Dntp/ddNTP, suivie du réassemblage et de l'amplification. L'intérêt de ce procédé visant à améliorer la structure et/ou la fonction protéiques est démontré par la création de nouveaux variants de sarcosine oxydase et la recombinaison de phosphatase alcaline placentaire humaine et de phosphatase alcaline intestinale du veau pour de meilleurs mutants présentant de meilleures propriétés.
