A method for obtaining a cRNA for use in generating calibration data, e.g., a standard curve, for absolute quantitation of RNA by RT-PCR is disclosed. The method includes the steps of: providing a synthetic oligonucleotide comprising an amplicon, a promoter sequence located 3' relative to the amplicon; synthesizing complementary RNA (cRNA) by in vitro transcription of the synthetic oligonucleotide; quantitatively assaying the cRNA by an independent method; and generating calibration data using a known quantity of the cRNA.

L'invention concerne un procédé d'obtention d'un ARNc utilisé dans l'établissement de données d'étalonnage, par exemple d'une courbe d'étalonnage, pour la quantification absolue d'ARN par RT-PCR. Le procédé comprend les étapes suivantes : obtention d'un oligonucléotide synthétique comprenant un amplicon, une séquence promoteur disposée en 3' par rapport à l'amplicon ; synthétisation complémentaire d'ARN (ARNc) par transcription in vitro de l'oligonucléotide synthétique ; dosage quantitatif d'ARNc par une méthode indépendante ; et établissement des données d'étalonnage au moyen d'une quantité connue d'ARNc.
