The present invention relates to methods of making activated linkers, particularly activated linkers having reactive Michael acceptors, and to an efficient process for purifying the activated linkers which utilizes hydrophobic interaction chromatography (HIC) to separate the activated linkers based on their size and on their end-group functionality. The purified activated linkers can be used to modify biologically active molecules and improve overall production of such molecules.
