A method for detecting interactions between first and second interacting molecules at variable sensitivity. This variable sensitivity may be obtained by providing for the overexpression of either a bait hybrid protein containing a DNA binding domain (desensitization) or a prey hybrid protein containing the DNA activation domain for a reporter gene (enhanced sensitivity). The use of exogenous activators of one or the other according to the needs of a particular system is readily accomplished.

L'invention porte sur un procédé de détection d'interactions entre une première et une deuxième molécule interagissant avec une sensibilité variable laquelle peut s'obtenir en provoquant la surexpression de soit une protéine hybride appât contenant un domaine de fixation d'ADN (désensibilisation), soit une protéine hybride proie contenant le domaine d'activation de l'ADN d'un reporter (sensibilité accrue). On peut facilement utiliser les activateurs exogènes de l'une ou de l'autre selon les besoins d'un système particulier.
