An adjuvant composition which is a stable oil-in-water emulsion comprising a metabolizable oil, one or more surfactants, an antioxidant and a compound to make the emulsion isotonic is described and claimed. The stable emulsion has a hydrophobic-lipophilic balance (HLB) of from about 7.5 to about 10.5 and a particle size of less than 3 µm. In a preferred embodiment, the stable emulsion comprises 10 % volume to volume squalene, 0.09 % weight to volume PLURONIC F-68 block co-polymer, 1.9 % weight to volume egg phosphatidyl choline, 1.75 % volume to volume glycerol and 0.05 % weight to volume .alpha. tocopherol. The preferred emulsion has a HLB of 8.0 and a particle size of about 0.2 µm. In a particularly preferred embodiment, the stable emulsion is combined with an attenuated lipid A derivative such as monophosphoryl lipid A or 3-deacylated monophosphoryl lipid A to enhance the adjuvanticity of the composition.
L'invention concerne une composition d'adjuvant qui constitue une émulsion huile dans l'eau stable renfermant une huile métabolisable, un ou plusieurs tensioactifs, un antioxydant et un composé servant à rendre l'émulsion isotonique. L'émulsion stable présente un équilibre hydrophobe-lipophile d'environ 7,5 à environ 10,5, et une taille des particules inférieure à 3 µm. Dans un mode de réalisation préféré, l'émulsion stable comporte 10 % volume à volume de squalène, 0,09 % poids à volume de copolymère séquencé PLURONIC F-68, 1,9 % poids à volume de phosphatidylcholine d'oeuf, 1,75 % volume à volume de glycérol et 0,05 % poids à volume d'.alpha.-tocophérol. L'émulsion préférée présente un équilibre hydrophobe-lipophile de 8,0 et une taille des particules d'environ 0,2 µm. Dans un mode de réalisation particulièrement préféré, l'émulsion stable est combinée à un dérivé de lipide A atténué tel qu'un monophosphoryl lipide A ou un monophosphoryl lipide A 3-désacétylé pour améliorer les propriétés d'adjuvant de la composition.
