A method for protein identification in complex mixtures that utilizes affinity selection of constituent proteolytic peptide fragments unique to a protein analyte. These "signature peptides" function as analytical surrogates. Mass spectrometric analysis of the proteolyzed mixture permits identification of a protein in a complex sample without purifying the protein or obtaining its composite peptide signature.

La présente invention concerne un procédé d'identification de protéines dans des mélanges complexes, faisant intervenir l'utilisation de l'affinité sélective de fragments de peptide protéolytique de constituant, caractéristiques d'un analyte protéique. Ces "peptides à signature" agissent comme des substituts analytiques. Une analyse par spectroscopie de masse du mélange protéolysé permet l'identification d'une protéine dans un échantillon complexe, sans faire appel ni à la purification de la protéine, ni à l'obtention de sa signature peptidique composée.
