A therapeutic method of modulating the immune response, by administering to a patient an amount of IL-4 effective to promote peripheral blood lymphocyte adhesion to microvascular endothelial cells in lymphoid organs. The IL-4 is preferably coadministered with IL-1.beta.. An improved method of screening a cell line for the production of a binding partner that binds with a cell adhesion molecule by contacting the binding partner with IL4-activated and nonactivated microvascular endothelial cells, and selecting binding partners that bind to the IL4-activated microvascular endothelial cells but not to the nonactivated microvascular endothelial cells. The selected binding partners may thereafter be tested for the ability to block lymphocyte binding to cytokine-activated endothelial cells. The binding partners are preferably also characterized by binding to human VCAM-1 and to IL4- or TNF.alpha.-activated bone marrow stromal cells. A representative embodiments is mAb 6G10 produced by hybridoma ATTC No. HB 10519. A therapeutic method of modulating the immune response in a patient, by administering to the patient an agent that specifically binds to IL4-activated microvascular endothelial cells, in an amount effective to impede transmigration of lymphocytes that specifically bind to VCAM-1 from blood across postcapillary venules.

Méthode thérapeutique permettant de moduler la réponse immunitaire en administrant à un patient une quantité d'IL-4 servant à promouvoir l'adhésion des lymphocytes du sang périphérique aux cellules endothéliales microvasculaires dans les organes lymphoïdes. L'IL-4 est administré de préférence en même temps que l'IL-1-bêta. L'invention a trait à une méthode améliorée de triage d'une lignée cellulaire pour la production d'un partenaire de liaison qui se lie à une molécule d'adhésion cellulaire; elle consiste à mettre le partenaire de liaison en contact avec des cellules endothéliales microvasculaires activées par IL-4 et non activées et à sélectionner des partenaires de liaison qui se fixent à des cellules endothéliales microvasculaires activées par IL-4 mais non à des cellules endothéliales microvasculaires non activées. On peut ensuite vérifier si les partenaires de liaison sélectionnés peuvent bloquer la liaison des lymphocytes aux cellules endothéliales activées par la cytokine. De préférence, les partenaires de liaison se lient également à VCAM-1 d'origine humaine et aux cellules du stroma de la moelle osseuse activées par IL-4 ou TNF-alpha. Un exemple représentatif est mAb 6G10 produit par l'hybridome ATTC no HB 10519. Méthode thérapeutique permettant de moduler la réponse immunitaire d'un patient en lui administrant un agent qui se lie spécifiquement aux cellules endothéliales microvasculaires activées par IL-4 à une concentration pouvant empêcher la transmigration de lymphocytes qui se lient spécifiquement à VCAM-1 dans le sang de veinules post-capillaires.
