Disclosed are methods for identifying an agent that modulates an immune response. One such method comprises screening for agents which inhibit the interaction between a PD-1 ligand and a PD-1 polypeptide, and determining whether the agents inhibit the interaction between a PD-1 ligand and a B7 polypeptide, to identify an agent that inhibits PD-1 ligand and PD-1 polypeptide interaction, while not inhibiting the interaction between a PD-1 ligand and a B7 polypeptide, as an agent that modulates an immune response. Another such method comprises screening for agents which inhibit the interaction between a PD-1 ligand and a B7 polypeptide, and determining whether the agents inhibit the interaction between a PD-1 ligand and a PD-1 polypeptide, to identify an agent that inhibits the PD-1 ligand and B7 polypeptide interaction, which does not inhibit the interaction between a PD-1 ligand and a PD-1 polypeptide, as an agent that modulates the immune response. Further disclosed is a method for inhibiting the interaction between a B7 polypeptide and a PD-1 ligand, the method comprising contacting an immune cell bearing a PD-1 ligand, or an immune cell bearing a B7 polypeptide, with an agent that inhibits the interaction between the PD-1 ligand and the B7 polypeptide. Such agents may be an anti- PD-1 ligand antibody or a small molecule. Also disclosed is a method for modulating an immune response. The method comprises contacting an immune cell bearing the PD-1 ligand, or an immune cell bearing the PD-1 polypeptide, with an agent that inhibits interactions between the PD-1 ligand and the PD-1 polypeptide, without inhibiting interactions between the PD-1 ligand and a B7 polypeptide, to thereby modulate an immune response. The agent may be an anti- PD-1 ligand antibody or a small molecule.

L'invention concerne des procédé d'identification d'agent modulant une réponse immune. Un tel procédé consiste à réaliser un criblage d'agents qui inhibent l'interaction entre un ligand PD-1 et un polypeptide PD-1, et à déterminer si les agents inhibent l'interaction entre un ligand PD-1 et un polypeptide B7, à identifier un agent inhibant une interaction entre un ligand PD-1 et un polypeptide PD-1, sans inhibition d'interaction entre un ligand PD-1 et un polypeptide B7, en tant qu'agent modulant une réponse immune. Un autre procédé consiste à réaliser un criblage d'agents qui inhibent l'interaction entre un ligand PD-1 et un polypeptide B7, et à déterminer si les agents inhibent l'interaction entre un ligand PD-1 et un polypeptide PD-1, afin d'identifier un agent qui inhibe l'interaction entre un ligand PD-1 et un polypeptide B7, sans inhiber l'interaction entre un ligand PD-1 et un polypeptide PD-1, en tant qu'agent modulant la réponse immune. L'invention concerne aussi un procédé d'inhibition de l'interaction entre un polypeptide B7 et un ligand PD-1, ce procédé consistant à mettre en contact une cellule immune portant un ligand PD-1, ou un polypeptide B7, avec un agent qui inhibe l'interaction entre le ligand PD-1 et le polypeptide B7. Ces agents peuvent être représentés par un anticorps ligand anti-PD-1 ou par une petite molécule. L'invention concerne enfin un procédé de modulation de réponse immune. Ce procédé consiste à mettre en contact une cellule immune portant le ligand PD-1, ou le polypeptide PD-1, avec un agent qui inhibe les interactions entre le ligand PD-1 et le polypeptide PD-1, sans inhiber les interactions entre le ligand PD-1 et un polypeptide B7, ce qui permet la modulation d'une réponse immune. L'agent peut être représenté par un anticorps ligand anti- PD-1 ou par une petite molécule.
