The invention features compositions and methods for site-specific modification of proteins by incorporation of an aldehyde tag. Enzymatic modification at a sulfatase motif of the aldehyde tag through action of a formylglycine generating enzyme (FGE) generates a formylglycine (FGly) residue. The aldehyde moiety of FGly residue can be exploited as a chemical handle for site-specific attachment of a moiety of interest to a polypeptide.

La présente invention concerne des compositions et des procédés utilisés dans le cadre d'une modification de protéines en un site spécifique, par introduction de marqueurs d'aldéhyde. Une modification enzymatique au niveau d'un motif sulfatase du marqueur d'aldéhyde sous l'effet d'une enzyme générant la formylglycine (FGE) produit un résidu formylglycine (FGly). Le groupe aldéhyde du résidu FGly peut être utilisé comme une poignée chimique pour assurer une liaison à un site spécifique d'un groupe cible sur un polypeptide.
