The present invention relates to methods of producing an allelic series of modifications in genes of interest in a cell. In particular, the invention provides methods for using nucleic acid sequence-modifying agents (e.g. chemicals, electromagnetic radiation, etc.) to introduce modifications in any nucleic acid sequence in the genome of a cell. Also provided are sets of cells which contain at least one modification in any gene of interest. The methods and compositions of the invention are useful in determining the function of the gene of interest.
L'invention concerne des procédés de fabrication d'une série allélique de modifications dans les gènes d'intérêt d'une cellule. L'invention concerne notamment des procédés d'utilisation d'agents modifiant la séquence d'acides nucléiques (p.ex., des produits chimiques, des rayonnements électromagnétiques, etc.) pour apporter des modifications dans n'importe quelle séquence d'acides nucléiques du génome d'une cellule. L'invention concerne en outre des ensembles de cellules qui comportent au moins une modification dans n'importe quel gène d'intérêt. Les procédés et les compositions de l'invention sont utiles pour déterminer la fonction d'un gène d'intérêt.
