Compounds of formula (I) have activity in alleviating the effects of impaired dopaminergic signaling, for example in the treatment of Parkinsons Disease: formula (I) wherein: R1 is a carboxyl, carboxyl ester, or carboxamide group; R2 and R3 are independently hydrogen, or a group -C(=O)R6 or -C(=O)OR6 wherein R6 is C1-C6 alkyl, or a group -CH2Q wherein Q is an optionally substituted monocyclic carbocyclic or heterocyclyl ring of 3 to 6 ring atoms; R4 and R5are independently (a) the side chain of a natural amino acid, or (b) optionally substituted C1-C4 alkyl, C2-C4 alkenyl, or C2-C4 alkynyl, or (c) -CH2XCH3, - CH2CH2XCH3, or -CH2XCH2CH3, wherein X is -O-, S, or -NR7 wherein R7 is hydrogen, methyl or ethyl; or (d) -CH2Q or CH2OQ wherein Q is as defined in relation to R6; or R4 and R5 taken together with the carbon atom to which they are attached form an optionally substituted cycloalkyl or heterocyclic ring of 3 to 8 ring atoms, optionally fused to a second, optionally substituted, carbocyclic or heterocyclic ring.
L'invention concerne des composés de formule (I) ayant une activité de soulagement des effets d'un signalement dopaminergique déficient, par exemple pour le traitement de la maladie de Parkinson. Formule (I) dans laquelle : R1 représente un carboxyle, un ester de carboxyle ou un groupement carboxamide ; R2 et R3 représentent indépendamment l'hydrogène ou un groupement -C(=O)R6 ou -C(=O)OR6 dans lequel R6 représente C1-C6 alkyle, ou un groupement -CH2Q dans lequel Q représente un cycle monocyclique, carbocyclique ou hétérocyclique éventuellement substitué ayant 3 à 6 atomes cycliques ; R4 et R5 représentent indépendamment (a) la chaîne latérale d'un acide aminé naturel ou (b) un C1-C4 alkyle éventuellement substitué, un C2-C4 alcényle, ou un C2-C4 alcynyle, ou (c) -CH2XCH3, -CH2CH2XCH3 ou -CH2XCH2CH3, X représentant -O-, S, ou -NR7, R7 représentant l'hydrogène, un méthyle ou un éthyle ; ou (d) -CH2Q ou CH2OQ, Q étant tel que défini pour R6 ; ou R4 et R5 forment avec l'atome de carbone auquel ils sont attachés un cycle cycloalkyle ou hétérocyclique éventuellement substitué ayant 3 à 8 atomes cycliques, éventuellement condensé avec un second cycle carbocyclique ou hétérocyclique éventuellement substitué.
