A compound of formula V or formula V1 for use in the treatment of a demyelinating peripheral neuropathy: wherein X is O, S, SO or SO2, R1 is halogen, trihalomethyl, OH, C1-7alkyl, C1- 4alkoxy, trifluoromethoxy, phenoxy, cyclohexylmethyloxy, pyridylmethoxy, cinnamyloxy, naphthylmethoxy, phenoxymethyl, CH2-OH, CH2-CH2- OH, C1-4alkylthio, C1-4alkylsulfinyl, C1-4alkylsulfonyl, benzylthio, acetyl, nitro or cyano, or phenyl, phenylC1- 4alkyl or phenyl-C1-4alkoxy each phenyl group thereof being optionally substituted by halogen, CF3, C1-4alkyl or C1-4alkoxy; R2 is H, halogen, trihalomethyl, C1-4alkoxy, C1-7alkyl, phenethyl or benzyloxy; R3 is H, halogen, CF3, OH, C1-7alkyl, C1-4alkoxy, benzyloxy, phenyl or CMalkoxymethyl; each of R4 and R5, independently is H or a residue of formula (a),wherein each of R8 and R9, independently, is H or C1-4alkyl optionally substituted by halogen; and n is an integer from 1 to 4; and the N-oxide derivatives thereof or prodrugs thereof, or a pharmaceutically acceptable salt, solvate or hydrate thereof, wherein R1a is halogen, trihalomethyl, C1-4alkyl, C1-4alkoxy, C1-4alkylthio, C1-4alkyl--sulifinyl, Cl-4alkyl-sulfonyl, aralkyl, optionally substituted phenoxy or aralkyloxy; R2a is H, halogen, trihalomethyl, C1-4alkyl, C1-4alkoxy, aralkyl or aralkyloxy; R3a is H, halogen, CF3, C1-4alkyl, C1- 4alkoxy, C1-4alkylthio or benzyloxy; R4a is H, C1-4alkyl, phenyl, optionally substituted benzyl or benzoyl, or lower aliphatic C1-5acyl; R5a is H, monohalomethyl, C1-4alkyl, C1-4alkoxy-methyl, C1-4alkyl-thiomethyl, hydroxyethyl, hydroxypropyl, phenyl, araikyl, C2- 4alkenyl or -alkynyl; R6a is H or C1-4alkyl; R7a is H, C1-4alkyl or a residue of formula (a) as defined above, X3 is O, S, SO or SO2; n a is an integer of 1 to 4; and designates a chiral centre of (R) or (S) configuration and the formula includes racemic and other mixtures of (R) and (S) configuration molecules; and the N-oxide derivatives thereof or prodrugs thereof, or a pharmaceutically acceptable salt, solvate or hydrate thereof.
La présente invention concerne un composé de formule V ou de formule VI en vue dune utilisation dans le traitement dune neuropathie périphérique démyélinisante : dans lesquelles X représente O, S, SO ou SO2, R1 représente un atome dhalogène, un groupe trihalogénométhyle, OH, alkyle en C1 à C7, alcoxy en C1 à C4, trifluorométhoxy, phénoxy, cyclohexylméthyloxy, pyridylméthoxy, cinnamyloxy, naphtylméthoxy, phénoxyméthyle, CH2-OH, CH2-CH2-OH, alkyl en C1 à C4-thio, alkyl en C1 à C4-sulfinyle, alkyl en C1 à C4-sulfonyle, benzylthio, acétyle, nitro ou cyano ou phényle, phénylalkyle en C1 à C4 ou phénylalcoxy en C1 à C4, chaque groupe phényle de ces groupes étant éventuellement substitué par un atome dhalogène, un groupe CF3, alkyle en C1 à C4 ou alcoxy en C1 à C4; R2 représente H, un atome dhalogène, un groupe trihalogénométhyle, alcoxy en C1 à C4, alkyle en C1 à C7, phénéthyle ou benzyloxy; R3 représente H, un atome dhalogène, CF3, OH, alkyle en C1 à C7, alcoxy en C1 à C4, benzyloxy, phényle ou CMalcoxyméthyle; chacun de R4 et R5, représente indépendamment H ou un résidu de formule (a), dans laquelle chacun de R8 et R9, représente indépendamment H ou un groupe alkyle en C1 à C4 éventuellement substitué par un atome dhalogène; et n est un nombre entier de 1 à 4; et leurs dérivés N-oxydes ou précurseurs, ou leurs sels pharmaceutiquement acceptables, solvates ou hydrates, dans lesquels R1a représente un atome dhalogène, un groupe trihalogénométhyle, alkyle en C1 à C4, alcoxy en C1 à C4, alkyl en en C1 à C4-thio, alkyl en C1 à C4-sulfinyle, alkyl en C1 à C4-sulfonyle, aralkyle, phénoxy ou aralkyloxy éventuellement substitué; R2a représente H, un atome dhalogène, un groupe trihalogénométhyle, alkyle en C1 à C4, alcoxy en C1 à C4, aralkyle ou aralkyloxy; R3a représente H, un atome dhalogène, un groupe CF3, CMalkyle, alcoxy en C1 à C4, alkyl en C1 à C4-thio ou benzyloxy; R4a représente H, alkyle en C1 à C4, phényle, benzyle ou benzoyle éventuellement substitué, ou acyle en C1 à C5 aliphatique inférieur; R5a représente H, un groupe monohalogénométhyle, alkyle en C1 à C4, alcoxy en C1 à C4-méthyle, alkyl en C1 à C4-thiométhyle, hydroxyéthyle, hydroxypropyle, phényle, aralkyle, alcényle ou alcynyle en C2 à C4; R6a représente H ou alkyle en C1 à C4; R7a représente H, un groupe alkyle en C1 à C4 ou un résidu de formule (a) tel que défini ci-dessus, X3 représente O, S, SO ou SO2; na est un nombre entier de 1 à 4; et * désigne un centre chiral de configuration (R) ou (S) et la formule comprend des mélanges racémiques et autres de molécules de configuration (R) et (S); et leurs dérivés N-oxydes ou précurseurs, ou leurs sels pharmaceutiquement acceptables, solvates ou hydrates.
