A class of substituted aminocyclohexane derivatives are selective agonists of 5-HT1-like receptors, being potent agonists of the human 5-HT1D.alpha. receptor subtype whilst possessing at least a 10-fold selective affinity for the 5-HT1D.alpha. receptor subtype relative to the 5-HT1D.beta. subtype; they are therefore useful in the treatment and/or prevention of clinical conditions, in particular migraine and associated disorders, for which a subtype-selective agonist of 5-HT1D receptors is indicated, whilst eliciting fewer side-effects, notably adverse cardiovascular events, than those associated with non-subtype-selective 5-HT1D receptor agonists.
La présente invention concerne une classe de dérivés aminocyclohexane substitués qui constituent des agonistes de récepteurs du type 5-HT¿1?. En effet, ce sont de puissants agonistes du sous-type de récepteur 5-HT¿1D.alpha.? humain qui présentent en outre une affinité 10 fois plus sélective pour le sous-type de récepteur 5-HT¿1D.alpha.? par comparaison au sous-type 5-HT¿1D.beta.?. Ces dérivés conviennent donc particulièrement au traitement et/ou la prévention d'états cliniques, en particulier les migraines et troubles associés, pour lesquels un agoniste des récepteurs 5-HT¿1D? à sélectivité de sous-type est indiqué, et ce, tout en entraînant moins d'effets secondaires, principalement les incidents cardio-vasculaires, qu'avec les agonistes des récepteurs 5-HT¿1D? ne présentant pas de sélectivité de sous-type.
