Provided is a compound that is an NMDA receptor antagonist having a broader safety margin, and is useful as an agent for treating or preventing Alzheimer's disease, cerebrovascular dementia, Parkinson's disease, ischemic apoplexy, or pain. A novel compound or salt thereof represented by formula I (see formula I) which is characterized in that is has an amino group and R1 (lower alkyl, cycloalkyl, -lower alkylene- aryl, aryl which may be substituted, and the like) on carbon atoms of indane, cyclopenta[b]thiophene, cyclopenta[b]furan, cyclopenta[b]pyridine, or cyclopenta[c]pyridine ring, or a 2,3-dihyro-1-benzofuran, 2,3-dihydro-1-benzothiophene or indoline ring, or the like, and has R2 and R3 (the same or different, each lower alkyl or aryl) on carbon atoms beside them, and an NMDA receptor antagonist comprising the same as an active ingredient.
L'invention concerne un composé qui puisse agir comme antagoniste de NMDA ayant un large intervalle de sécurité et qui soit utile comme agent thérapeutique ou comme agent prophylactique pour la maladie d'Alzheimer, la démence cérébrovasculaire, la maladie de Parkinson, l'accident ischémique cérébral ou une douleur. L'invention concerne donc un nouveau composé caractérisé par le fait qu'il présente un groupe amino et un groupe R1 [qui représente un alkyle inférieur, un cycloalkyl, alkylène inférieur-aryle, un aryle qui peut avoir un substituant ou autre substituant] sur un atome de carbone dans un anneau indane, cyclopenta[b]thiophène, cyclopenta[b]furane, cyclopenta[b]pyridine ou cyclopenta[c]pyridine ou un anneau 2,3-dihydro-1-benzofurane, 2,3-dihydro-1-benzothiophène ou indoline ou autre anneau, et ayant également des groupes R2 et R3 [qui représentent indépendamment un alkyle inférieur ou un aryle] sur un atome de carbone adjacent à l'atome de carbone mentionné ci-dessus, ou un sel de celui-ci ; et un antagoniste de NMDA comprenant le composé ou le sel comme ingrédient actif.
