Methods and vectors (both DNA and retroviral) are provided for the construction of a Library of mutated cells. The Library will preferably contain mutations in essentially all genes present in the genome of the cells. The nature of the Library and the vectors allow for methods of screening for mutations in specific genes, and for gathering nucleotide sequence data from each mutated gene to provide a database of tagged gene sequences. Such a database provides a means to access the individual mutant cell clones contained in the Library. The invention includes the described Library, methods of making the same, and vectors used to construct the Library. Methods are also provided for accessing individual parts of the Library either by sequence or by pooling and screening. The invention also provides for the generation of non-human transgenic animals which are mutant for specific genes as isolated and generated from the cells of the Library.
Procédés et vecteurs (ADN et rétrovirus) pour l'élaboration d'une banque de cellules mutantes, contenant de préférence des mutations qui concernent essentiellement tous les gènes présents dans le génome des cellules. La nature de la banque et des vecteurs permet l'utilisation de procédés de criblage pour la recherche de mutations relatives à des gènes spécifiques, et des procédés de collecte des données relatives à la séquence nucléotidique de chaque gène mutant, dans le but d'élaborer une base de données sur les séquences géniques marquées. Cette base de données permet d'accéder individuellement aux clones des cellules mutantes de la banque. On décrit par ailleurs la banque de l'invention, un certain nombre de procédés relatifs à son élaboration et différents vecteurs utilisés dans ladite élaboration. On décrit enfin des procédés pour accéder aux éléments individuels de la banque, par séquence ou encore par groupement et criblage, et aussi la constitution d'animaux transgéniques non humains qui sont mutants pour un certain nombre de gènes spécifiques tels qu'isolés et générés à partir des cellules de la banque.
