The invention relates to a novel retrotransposon isolated from the yeast Candida albicans. The new retrotransposon, designated pCal, belongs to the Ty1/Copia group. The free, linear, double stranded DNA form of the element is highly expressed such that it can seen as a distinct band when uncut genomic C.albicans DNA is analysed on a agrose gel. The retrotransposon has a copy number of between 40 and 150 DNA copies of itself per genome and may be used in a method of introducing DNA into the genome of a cell. It may also be used in a gene disruption and/or gene discovery system.
