An apparatus and method for analysing temperature-dependent molecular configurations such as folding comprises a multi-channel flow-through chip (12) along which molecules to be analysed pass. A temperature gradient is maintained along the length of the chip. As molecules pass along the channels they fold or unfold, in response to the changing temperature. These changing molecular configurations are investigated by simultaneously measuring the extent to which the molecules absorb UV light, and the extent to which they fluoresce. The absorption and fluorescence information is supplied to a computer system (26) for real-time analysis.
L'invention porte sur un appareil et un procédé d'analyse de configurations de molécules dépendant de la température telles que le pliage utilisant une puce (12) à flux traversant à plusieurs canaux le long desquels passent les molécules à analyser, un gradient de température étant maintenu sur toute la longueur de la puce. Alors que les molécules défilent dans les canaux, elles se plient ou se déplient en fonction des variations de température. Ces variations de configuration sont analysées par des mesures simultanées de leur niveau d'absorption des UV, et de leur niveau de fluorescence. Les informations d'absorption et de fluorescence sont fournies à un ordinateur (26) en vue de leur analyse en temps réel.
