A method in and a kit for analytical methods in a flow matrix, which methods utilize biospecific affinity reactions to detect an analyte in a sample. The method and kit, respectively, are based on allowing the sample (analyte) and an analytically detectable reactant (Reactant*) to migrate through flow channels in a flow matrix to a detection zone (DZ) located in the matrix, in which zone there is a firmly anchored biospecific affinity reactant (Capturer), the Reactant* being captured in the detection zone (DZ) in an amount related to the amount of analyte in the sample. The method and kit are characterized in that A) Reactant* has particles as analytically detectable group, and B) the Capturer is anchored to the matrix via immobilized particles, which preferably are smaller than the smallest inner dimension of the flow channels and which preferably on their surface exhibit hydrophilic groups.
On décrit des procédés et une trousse d'analyse qui, dans une matrice d'écoulement, mettent en oeuvre des réactions d'affinité biospécifique pour déterminer un analysat dans un échantillon. Ces procédés et cette trousse ont pour rôle respectif de faire migrer un échantillon (analysat) et un réactif décelable par analyse (réactif*) à travers des voies d'écoulement, vers une zone de détection (DZ) située dans dans la matrice d'écoulement, ladite zone de détection contenant un réactif d'affinité biospécifique (agent de capture) solidement fixé. Le réactif* est capturée dans la zone de détection (DZ) en quantité liée à la quantité d'analysat dans l'échantillon. Les procédés et la trousse se cractérisent en ce que a) le réactif* comprend des particule en tant que groupe décelable par analyse, et b) l'agent de capture est fixé à la matrice par l'intermédiaire de particules immobilisées qui, de préférence, sont plus petites que la plus petite dimension interne des voies d'écoulement et présentent à leur surface des groupes hydrophiles d'exposition.
