This invention relates to a process for the synthesis of ketomethylene derivatives of the tripeptide Phe-Gly-Pro ("keto-ACE", compound 5a) and analogues thereof. The synthesis process proceeds via an .alpha.,~-unsaturated keto intermediate. A key feature of the process involves a Horner-Emmons olefination of the , -unsaturated keto-phoshonate with ethyl gyloxylate. Keto- ACE analogues produced by the process of the invention display C-domain selectivity.
Cette invention concerne un procédé pour la synthèse de dérivés cétométhylène du tripeptide Phe-Gly-Pro (enzyme céto-ACE, composé 5a) et d'analogues de celui-ci. Ce procédé de synthèse s'effectue via un intermédiaire céto .alpha.,.beta.-insaturé. Une caractéristique clé de ce procédé implique une oléfination Horner-Emmons du céto-phoshonate .alpha., .beta.-insaturé par de l'éthyle-gyloxylate. Des analogues de l'enzyme céto-ACE produits par ce procédé présentent une sélectivité du domaine C.
